DNA reparasjon og adaptiv immunitet

Hvordan dannes og hvordan repareres skader i arvestoffet vårt?  Hvordan benytter immunforsvaret samme molekylære mekanismer for å generere antistoffer til bekjempelse av infeksjon?  Hva går galt når friske celler akkumulerer mutasjoner og omdannes til kreftceller?  Dette er viktige spørsmål i min forskning.

Sentralt i  forskningen er DNA-skaden uracil. I motsetning til i RNA, er ikke uracil en normal base i DNA, men dannes ved spontan eller enzymatisk fjerning av aminogruppen på cytosin. Alle levende organismer har enzymer som kan fjerne uracil fra DNA, såkalte uracil-DNA glycosylaser. Disse er viktige for å forhindre mutasjoner. Karakterisering av disse enzymene har blant annet resultert i den første publiserte proteinstrukturen til et DNA-reparasjonsenzym fra menneske. I tillegg la det grunnlag for den molekylære forståelsen av hvordan et enzym kan gjenkjenne og dytte ut en modifisert base bland alle de normale basene i DNA helixen.

I den senere tid har det blitt klart at immunforsvaret benytter seg av enzymer som spesifikt introduserer uracil i DNA (DNA deaminaser). Dette benyttes både for å inaktivere virus DNA (APOBEC enzymer) og for å generere nye antistoffer ved somatisk hypermutasjon (SHM) og klasse switch rekombinasjon (CSR). I aktiverte B-celler utrykkes activation-induced cytidine deaminase (AID). AID deaminerer cytosin til uracil i immunoglobulin (Ig) genene, noe som initierer både SHM og CSR. Uracil-DNA glycosylase er også sentrale for disse nøkkelprosessene i vårt adaptive immunforsvar.

AID er et mutatorprotein som er veldig sentralt i utviklingen av blant annet B-celle lymfom.  Uregulert AID aktivitet kan også mutere gener som er viktige ved utvikling av kreft. I den senere tid har regulering av AID og AID-generert genomisk uracil i forhold til genomstabilitet og utvikling av kreft vært mitt viktigste forskningsfokus.
